  • ntroduction: Research on DNA analysis of ancient human teeth is still not optimal due to the lack of the DNA
    extraction process. Although teeth are the most resistant part of the body to protect the organic and inorganic
    materials, problems in extracting ancient human DNA still occur which results in short DNA molecules which
    can complicate the DNA extracting process. This research was conducted to map the success of ancient human
    teeth DNA extraction using a variety of substrates that were successfully carried out by previous researchers
    Methods: This research was conducted using literature review method through a rapid review approach by
    collecting articles through an electronic search engine such as PubMed, EbscoHost, Sicence Direct, Research
    Gate, and Cambridge Core that matched the inclusion and exclusion criteria. Results: From 6 articles that
    included, the ancient human DNA extraction method with silica-based materials was more often used by
    researcher, especially for short DNA base pairs. Conclusion: Extracting ancient human tooth DNA using a silicabased protocol with poor preservation can produced historical age, mtDNA, and microbiota in dental calculus in
    fully demineralized samples. Meanwhile, extracting DNA from ancient human teeth using phenol-chloroform can
    describe eyes and hair colour of ancient humans.
